Definitely - I've driven that same route and it's very nice. Shouldn't be too crowded since I would think most the leaves have already fallen.
EDIT: Looking at the Blue Ridge Parkway webcams, it doesn't appear as if the leaves have fallen so yes, Deja_Vu is probably right. I would expect some crowds.
